ABSTRACT

OBJECTIVE@#To investigate the effects of CACNA1H gene knockout (KO) on autistic-like behaviors and the morphology of hippocampal neurons in mice.@*METHODS@#In the study, 25 CACNA1H KO mice of 3-4 weeks old and C57BL/6 background were recruited as the experimental group, and 26 wild type (WT) mice of the same age and background were recruited as the control group. Three-chamber test and open field test were used to observe the social interaction, anxiety, and repetitive behaviors in mice. After that, their brain weight and size were measured, and the number of hippocampal neurons were observed by Nissl staining. Furthermore, the CACNA1H heterozygote mice were interbred with Thy1-GFP-O mice to generate CACNA1H-/--Thy1+(KO-GFP) and CACNA1H+/+-Thy1+ (WT-GFP) mice. The density and maturity of dendritic spines of hippocampal neurons were observed.@*RESULTS@#In the sociability test session of the three-chamber test, the KO mice spent more time in the chamber of the stranger mice than in the object one (F1, 14=95.086, P < 0.05; Post-Hoc: P < 0.05), without any significant difference for the explored preference index between the two groups (t=1.044, P>0.05). However, in the social novelty recognition test session, no difference was observed between the time of the KO mice spend in the chamber of new stranger mice and the stranger one (F1, 14=18.062, P < 0.05; Post-Hoc: P>0.05), and the explored preference index of the KO mice was less than that of the control group (t=2.390, P < 0.05). In the open field test, the KO mice spent less time in the center of the open field apparatus than the control group (t=2.503, P < 0.05), but the self-grooming time was significantly increased compared with the control group (t=-2.299, P < 0.05). Morphological results showed that the brain weight/body weight ratio (t=0.356, P>0.05) and brain size (t=-0.660, P>0.05) of the KO mice were not significantly different from those of the control group, but the number of neurons were significantly reduced in hippocampal dentate gyrus compared with the control group (t=2.323, P < 0.05). Moreover, the density of dendritic spine of dentate gyrus neurons in the KO-GFP mice was significantly increased compared with the control group (t=-2.374, P < 0.05), without any significant difference in spine maturity (t=-1.935, P>0.05).@*CONCLUSION@#CACNA1H KO mice represent autistic-like behavior, which may be related to the decrease in the number of neurons and the increase in the density of dendritic spine in the dentate gyrus.

ABSTRACT

Objective To explore the impact of psychological alignment and coping style on nurse′s job satisfaction. Methods A total of 389 clinical nurses were investigated by the Sense of Coherence Scale, the simplified Coping Style Questionnaire and Minnesota Satisfaction Scale. Results The scores of job satisfaction, psychological alignment, positive coping and pessimistic coping of clinical nurses was (72.91 ± 11.10), (56.97 ± 10.29), (23.16 ± 5.40) and (9.80 ± 4.03) points respectively. The predictors of job satisfaction were sense of control, sense of meaning and positive coping style. Conclusions It is suggested that nursing managers should pay close attention to nurses′psychological alignment and guide clinical nurses adopt positive coping style, and thereby to improve nurses′job satisfaction.

ABSTRACT

<p><b>OBJECTIVE</b>To analysis the early complications of tibial fracture and its related factors, and propose a solution.</p><p><b>METHODS</b>From December 2003 to December 2013,38 patients with early complications of tibial plateau fracture after operation were retrospectively analyzed. There were 35 males and 3 females, aged from 37 to 69 years old (averaged 42.3 years). According to Schatzker classification, 3 cases were classified as type II, 2 cases as type III, 2 cases as type IV, 19 cases as type V, 12 cases as type VI. The intervals between injury and operation ranged from 9 hours to 9 days, 26 cases within 3 days. Fifteen cases were treated with internal fixation of plates and 23 were treated by plate fixation and bone transplantation. Early complications included skin necrosis in 15 cases, infection in 6 cases, osteofascial compartment syndrome in 3 cases, common peroneal nerve injury in 2 cases, the superficial peroneal nerve injury in 3 cases, popliteal artery injury in 2 cases, loss of reduction in 7 cases.</p><p><b>RESULTS</b>The wound of 14 cases healed at the first stage and 24 cases healed delay. Hospitalization days ranged from 7 to 67 days (averaged 25.6 days). All patients were followed up for 12 to 36 months with an average of 16.4 months. The fracture healing time ranged from 3 to 9 months (averaged 6.9 months). According to Merchant knee function evaluation criteria, the results were excellent in 19 cases, good in 12, fair in 5 and poor in 2.</p><p><b>CONCLUSION</b>Early complications of tibial fracture after operation is closely associated with the severe fracture complexity and related with preoperative preparation, surgical timing, operation incision selection and surgical technique. Early detection and timely processing reduce damage.</p>

ABSTRACT

Objective To explore the association between serum neutrophil‐to‐lymphocyte ratio (NLR) and DR in T2DM patients . Methods A total of 128 T2DM patients were recruited and divided into three groups :PDR group (n=22) ,NPDR group(n= 44) ,and simple T2DM group(n=62) .The levels of FPG ,HbA1 c ,TC ,TG and NLR were measured in each group . Results Spearman correlation analysis revealed that DR was positively correlated with T 2DM duration ,SBP ,DBP ,FPG ,HbA1 c ,TC , TG ,LDL‐C ,NLR ,CKD (r= 0.524 ,0.277 ,0.279 ,0.194 ,0.271 ,0.208 ,0.223 ,0.358 ,0.621 , 0.362 ,P<0 .05 or P<0 .01) .Logistic multiple regression analysis showed that ,T2DM duration ,NLR and CKD were independent risk factors for DR . Conclusion NLR level is associated with DR .

ABSTRACT

<p><b>OBJECTIVE</b>To evaluate the design principles, clinical results and significance of hatchet skin flaps for repairing tissue defects in different parts of cheek.</p><p><b>METHODS</b>The area of cheek was divided into three parts, P(I), P(II) and P(III), with vertical lines through the medial canthus and lateral canthus. Different kinds of hatchet skin flaps were designed to repair tissue defects in different part of cheek. The hatchet skin flaps were performed in 29 cases with tissue defects in different part of cheek from August 2005 to August 2009. There were 17 male and 12 female, aged from 19 to 81 years, with a mean age of (45 ± 16) years. The size of tissue defect ranged from 1.5 cm × 1.5 cm to 2.5 cm × 3.5 cm. Patients' satisfactions were evaluated with a questionnaire in 5 aspects:color and texture match, scar, morbidity, and function after 6 months operatively.</p><p><b>RESULTS</b>All the flaps survived completely with good color and tissue match. The facial contour was not altered obviously. Six to eighteen months later, all scars were almost invisible. All 29 patients were satisfied with their results.</p><p><b>CONCLUSIONS</b>The hatchet skin flap is one of the versatile reconstructive methods for repairing of medium and small defects in the three parts of cheek. Defects in different part of cheek should be repaired individually with hatchet flap based on characters of natural lines.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To evaluate the effect of tumescent infiltration solution temperature on core body temperature after liposuction.</p><p><b>METHODS</b>15 healthy female subjects were randomly divided into 2 groups to receive tumescent infiltration solution at 25 degrees C as group A, or at 37 degrees C as group B. All subjects were under epidural anesthesia. Vital signs, including core temperature, heart rate, respiratory rate and blood pressure, were monitored immediately, 1 hour, 2 hours, 3 hours, 4 hours and 8 hours after operation.</p><p><b>RESULTS</b>The core body temperature immediately, 1 hour, 2 hours and 3 hours after operation were (35.8 +/- 0.5) degrees C, (35.8 +/- 0.5) degrees C, (36.0 +/- 0.5) degrees C, (36.1 +/- 0.5) degrees C in group A, and (36.5 +/- 0.4) degrees C, (36.5 +/- 0.3) degrees C, (36.5 +/- 0.3) degrees C, (36.6 +/- 0.4) degrees C in group B, showing a significant difference between the two groups (P = 0.008, P = 0.008, P = 0.03, P = 0.033, respectively). There was no difference in body temperature 4 hours and 8 hours after operation and in heart rate, respiratory rate and blood pressure between the two groups (P > 0. 05).</p><p><b>CONCLUSIONS</b>The tumescent infiltration solutions at room temperature (25 degrees C) can decrease the core body temperature and increase surgical risk. It might not be good for rehabilitation. It is recommended to use tumescent infiltration solution at body temperature (37 degrees C) in liposuction.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To evaluate the therapeutic effect of hatchet flap for buccal tissue defect.</p><p><b>METHODS</b>The hatchet flap was designed beside the tissue defect and advanced to cover the defect.</p><p><b>RESULTS</b>Since 2006, 13 cases were treated with primary healing and no flap loss. The size of the flaps ranged from 1.8 cm x 2.0 cm to 2.5 cm x 3.5 cm. All the cases were followed up for 3 months to 1.5 years. The postoperative appearance was satisfactory with inconspicuous scar.</p><p><b>CONCLUSIONS</b>Hatchet flap is very suitable for the buccal tissue defect with satisfactory cosmetic result. The facial natural figure is protected very well.</p>

ABSTRACT

OBJECTIVE:To establish a HPLC method for content determination of forsythin in qingreling granules by C 18 solid phase extraction to purify the forsythin in qingreling granules.METHODS:The column of Dikma Diamonsil TM C 18 ,a mob_ ile phase of acetonitrile-water(30∶70),the flow rate of1.0ml/min,the detection wavelength of230nm,and the column temperature of30℃.RESULTS:The calibration curve of forsythin was linear in the range of10.0~200.0?g/ml(r=1.0000),the average recovery was98.1%(RSD=1.91%).CONCLUSION:The present method is accurate,reliable,simple and with good reproducibility,the sample was purified completely,and little interference and it can be used for quality control of qingreling granules.

ABSTRACT

Objective To study the chemical constituents of marine sponge Craniella australiensis collected from the South China Sea.Methods The compounds were isolated and purified by column chromatography and their structures were identified based on spectral analyses and chemical evidence.Results Four compounds were isolated from the ethanol extract and identified as: ?-sitosterol(Ⅰ);3?,6?-stigmast-4-en-3,6diol(Ⅱ);1,12-diazacyclodocosane-2,11-dione(Ⅲ);uracil(Ⅳ).Conclusion All these compounds were isolated from this sponge for the first time.
